X-Ray Absorption Spectromicroscopy of Aqueous Samples

X-ray PhotoElectron Emission Microscopy (X-PEEM) has found numerous applications in the surface microchemical analysis of materials science and biological specimens. We have reconfigured the MEPHISTO X-PEEM instrument, installed at the UW-Synchrotron Radiation Center, to measure true x-ray transmission spectra by converting transmitted photons to photoelectrons via a thin photocathode layer of gold. We have also developed a novel method of introducing aqueous samples into ultra high vacuum. Hence, for the first time, x-ray spectroscopy can be performed on biologically relevant elements (such as K, Ca, etc.) in a physiological environment, i.e. in solution. More importantly when coupled with X-PEEM imaging this technique may offer the unique and exciting possibility of studying living cells. We present the first x-ray transmission spectra of various solutions, including ionic and chelated Ca.
